In Lady of Burlesque Barbara Stanwyck plays Dixie Daisy (aka Deborah Hoople), a singer/dancer at a former opera house that has been transformed into a burlesque theater/review. Dixie is the new attraction at the theater, which invokes jealousy from some of the other female entertainers. Dixie immediately attracts the eye of Biff Brannigan (played by Michael O'Shea), who is also working at the theater as a comedy/dance man. Entertainers Dolly Baxter and Lolita La Verne engage in a very nasty dispute that turns ugly. Bitchy remarks are exchanged and a catfight ensues.

As Dixie and the other entertainers hoof their way through numerous song and dance routines a murderer is lurking around the theater waiting to strike. When one of the female entertainers is found strangled with Dixie's own g-string around her neck, Dixie is suddently a suspect. Dixie teams up with Biff to find the killer, who they believe is among them. Trouble continues to plague the theater when another female entertainer is found dead in one of the props that Dixie was scheduled to use for her act. Again, the not-so-swift detectives peg Dixie as their number one suspect. When Dixie spots the killer amongst the burlesque troop she uses herself as bait to catch a killer. As the murderer is identified and motives are established, Biff and Dixie dance off into the sunset.
